That's probably because of the symlink you added by hand, i.e. the one which
points to the brltty executable (/sbin/brltty on your system).

The S25 link is also pointing to the brltty executable. They're both wrong. I
suggest you remove them both and then try the update-rc.d command again.

I'm not personally familiar with the update-rc.d command (it's a
Debian-specific command), but I do have a note that the correct command line
is:

   update-rc.d brltty start 25 S .
