You need two different files. Use brltty's -f option to point to the one you'd 
like each of them to use. Run two different brlttys, like this:

   brltty -f /etc/brltty1.conf
   brltty -f /etc/brltty2.conf

Specify two devices, separated by a comma. Like this:

   braille-device serial:ttyS0,serial:ttyUSB0
