You need to tell BrlAPI which tty tmux is on. That isn't easy as tmux creates a pty (virtual console). That's why we wrote the brltty-tmux script. After starting tmux, rather than starting the second brltty instance directly, start it by running the brltty-tmux script. If you aren't running as root then use its -s option to tell it that it should internally use sudo.
